Phenol/Chloroform Extraction

1.Add equal volume of phenol chloroform to the solution you want to extract DNA from.

2.Vortex well.

3.Spin @ max speed for 2 min in cold room centrifuge

4.Remove aqueous layer. Thats the top layer. Be careful not to take any of the bottom layer.

Ethanol Precipitation

1.Add 10% 3M NaOAc pH 5.2 (this can be omitted)

2.Add 2.5 volumes 95% EtOH

3.Let sit at -20 C for 15.

4.Spin at max speed @4 C for 15 min

5.Aspirate (remove) 95% EtOH

6.Add ~1 volume of 70% EtOH.

7.Let sit at -20 C for 5min.

8.Spin @ max speed @4 C for 5 min

9.Aspirate (remove) 70% EtOH

10. Resuspend in sterile water approx 30-50 uL for a miniprep.
